The increasing use of plastic (synthetic polymers) results in the release of uncontrollable amounts of synthetic materials into the environment through waste, infrastructure, and essential goods. As plastic materials undergo weathering, a complex process unfolds, leading to the formation of pollutants, notably microplastics. This study employs multiple instrumental methods to explore the intricate abiotic degradation of the five most commonly used synthetic polymers in environmentally relevant conditions. An extensive set of analytical techniques, along with chemometric analysis of the results of Raman spectroscopy, was used to characterize the materials and evaluate the nature and extent of degradation caused by artificial weathering under temperature, humidity, and solar-like irradiation cycles. Investigation focuses on the link between abiotic weathering and the generation of micro- and nanoplastics, accompanied by molecular and surface adhesion changes, and the release of additives such as metals and metal oxides. Research reveals that microplastics may exhibit varied physical properties due to the incorporation of significant quantities of high-density additives from the parent plastic, which might influence the extraction methods and the transportation models' accuracy. At the molecular and microscopic scales, non-homogeneous pathways through which plastic decomposes during weathering were observed. The formation of additive-polymer combinations might play a pivotal role in the monitoring approaches for microplastics, presenting unique challenges in assessing the environmental impact of different plastic types. These findings offer complex insight into abiotic weathering, microplastics' generation, and the influence of additives that were previously overlooked in toxicity and health assessment studies. As plastic pollution continues to escalate, understanding these complex processes is crucial for microplastic monitoring development and adopting effective preventative measures.

In this review, recent advances in the methods of pre-treatment of plant material for the extraction of secondary metabolites with high biological activity are presented. The correct preparation of the material for extraction is as important as the selection of the extraction method. This step should prevent the degradation of bioactive compounds as well as the development of fungi and bacteria. Currently, the methods of preparation are expected to modify the particles of the plant material in such a way that will contribute to the release of bioactive compounds loosely bonded to cell wall polymers. This review presents a wide range of methods of preparing plant material, including drying, freeze-drying, convection drying, microwave vacuum drying, enzymatic processes, and fermentation. The influence of the particular methods on the structure of plant material particles, the level of preserved bioactive compounds, and the possibility of their release during the extraction were highlighted. The plant material pre-treatment techniques used were discussed with respect to the amount of compounds released during extraction as well their application in various industries interested in products with a high content of biologically active compounds, such as the pharmaceutical, cosmetics, and food industries.

Saponins are an important group of secondary metabolites naturally occurring in plants with important properties like: antibacterial, antiviral and antifungal. Moreover, they are widely used in the cosmetic industry and household chemistry. The sapogenins are saponin hydrolyses products, frequently used to facilitate saponin detection. In the present study, an improved methodology for isolation and separation of five sapogenins extracted from nettle (Urtica dioica L.), white dead-nettle (Lamium album L.), common soapwort (Saponaria officinalis L.) and washnut (Sapindus mukorossi Gaertn.) was developed using ultra-high-performance liquid chromatography with an evaporative light-scattering detector (UHPLC-ELSD). Based on quantitative analysis, the highest content of hederagenin (999.1 ± 6.3 µg/g) and oleanolic acid (386.5 ± 27.7 µg/g) was found in washnut extracts. Good recoveries (71% ± 6 up to 99% ± 8) were achieved for four investigated targets, while just 22.2% ± 0.5 was obtained for the fifth one. Moreover, hederagenin and oleanolic acid of whose highest amount was detected in washnut (999.1 ± 6.3 µg/g and 386.5 ± 27.7 µg/g, respectively) were subject to another approach. Consequently, liquid chromatography coupled mass spectrometry (LC/MS) with multiple reaction monitoring mode (MRM) was used as an additional technique for fast and simultaneous identification of the mentioned targets.

Cytokines play a huge role in many biological processes. Their production, release and interactions are subject to a very complex mechanism. Cytokines are produced by all types of cells, they function very differently and they are characterized by synergism in action, antagonism, and aggregation activity, opposing action of one cytokine, overlapping activity, induction of another cytokine, inhibition of cytokine synthesis at the mRNA level as well as autoregulation-stimulation or inhibition of own production. The predominance of pro-inflammatory cytokines leads to a systemic inflammatory response, and anti-inflammatory-to an anti-inflammatory response. They regulate the organism's immune response and protect it against sudden disturbances in homeostasis. The synthesis and activity of cytokines are influenced by the central nervous system through the endocrine system (pituitary gland, adrenal glands).

To elaborate a complete extraction protocol for the enhanced release of biologically active compounds from plant cells, this study aimed to optimize together the parameters of the supercritical fluid extraction (SFE) process (temperature, pressure, and percentage of cosolvent) and enzymatic treatment of plant material (pH, enzyme concentration, time, and temperature) by response surface methodology (RSM). Medicago sativa L. was selected as a plant material due to its richness in phenolics and flavonoids. HPLC-MS/MS analysis allowed evaluating the content of individual bioactive compounds in obtained extracts. The total content of polyphenolic compounds in the extract obtained after two-step optimization was much higher (546 ± 21 µg/g) than in the extract obtained from non-hydrolyzed material (275 ± 23 µg/g) and in the extract obtained by maceration (162 ± 20 µg/g). Furthermore, it was evidenced that extract with the highest content of polyphenolic compounds can support the cellular antioxidant system both as a free radical scavenger and by stimulating the antioxidant enzyme system.

Saponins in plant extracts were indirectly determined by estimation of the content of sapogenins. The first step of determination is extraction with high efficiency. One conventional extraction technique (maceration) and two modern ones (accelerated solvent extraction and supercritical fluid extraction) were compared. Methanol and ethanol were used as solvents or co-solvents. The results were supported by statistical analysis. Saponins were extracted from leaves, roots, and sprouts of Medicago sativa. Acid hydrolysis, purification, and determination by high-performance liquid chromatography with evaporative light scattering detector were used. The content of sapogenins was the highest in the roots. Smaller amounts of sapogenins were found in sprouts and the smallest ones in leaves. The main ingredient was medicagenic acid with mean concentration of 621.8 µg/g in roots, 456.7 µg/g in sprouts, and 471.3 µg/g in leaf extract. The highest content of sapogenins in extract was obtained after maceration with methanol; however, this method is nonselective in relation to biologically active compounds. Due to the possibility of using the obtained extracts with sapogenins in the cosmetic or pharmaceutical industry, the selection of extraction techniques and solvents is a very important aspect. Additionally, the chosen technique should be considered eco-friendly and consistent with the assumptions of "green chemistry."

Saponins are widespread secondary metabolites with various beneficial properties: fungicidal, antibacterial, antiviral, and anticancer. Alfalfa saponin molecules contain mainly: medicagenic acid, hederagenin, bayogenin, and soyasapogenol B. Structural diversity of saponins makes their determination in Medicago sativa extracts very difficult. The most popular determination technique is high-performance liquid chromatography applied with evaporative light scattering detection. Qualitative and quantitative analysis of sapogenins from Medicago sativa by high-performance liquid chromatography with evaporative light scattering detection required hydrolysis and purification of extracts obtained by supercritical fluid extraction. Hydrolysis of saponins with concentrated hydrochloric acid provided high concentration of medicagenic acid. In the purification process, satisfactory results were obtained for solid-phase extraction using octadecyl. Recoveries were from 71 to 99% with a standard deviation from 2 to 8. Hydrolysis with concentrated hydrochloric acid was the only method that allowed identification of all four analyzed sapogenins. Moreover, it is characterized by a short time of preparation, simplicity of execution, a small amount of the sample and solvents. The hydrolysis and purification methods coupled with high-performance liquid chromatography and evaporative light scattering detection can be successfully used for qualitative and quantitative analysis of the main saponins present in Medicago sativa plant extracts obtained by supercritical fluid extraction.

Based on the bioactive properties of certain compounds, such as antioxidant and anti-inflammatory activities, an interesting subject of research are natural substances present in various parts of plants. The choice of the most appropriate method for separation and quantification of biologically active compounds from plants and natural products is a crucial step of any analytical procedure. The aim of this review article is to present an overview of a comprehensive literature study from the last 10 years (2007-2017), where relevant articles exposed the latest trends and the most appropriate methods applicable for separation and quantification of biologically active compounds from plant material and natural products. Consequently, various extraction methods have been discussed, together with the available procedures for purification and pre-concentration and dedicated methods used for analysis.

Bottom sediments are very important in aquatic ecosystems. The sediments accumulate heavy metals and compounds belonging to the group of persistent organic pollutants. The accelerated solvent extraction (ASE) was used for extraction of 16 compounds from PAH group from bottom sediment of Vistula. For the matrix of candidate of a new reference material, moisture content, particle size, loss on ignition, pH, and total organic carbon were determined. A gas chromatograph with a selective mass detector (GC/MS) was used for the final analysis. The obtained recoveries were from 86% (SD=6.9) for anthracene to 119% (SD=5.4) for dibenzo(ah)anthracene. For the candidate for a new reference material, homogeneity and analytes content were determined using a validated method. The results are a very important part of the development and certification of a new reference materials.

Thermal performance curves for population growth rate r (a measure of fitness) were estimated over a wide range of temperature for three species: Coleps hirtus (Protista), Lecane inermis (Rotifera) and Aeolosoma hemprichi (Oligochaeta). We measured individual body size and examined if predictions for the temperature-size rule (TSR) were valid for different temperatures. All three organisms investigated follow the TSR, but only over a specific range between minimal and optimal temperatures, while maintenance at temperatures beyond this range showed the opposite pattern in these taxa. We consider minimal and optimal temperatures to be species-specific, and moreover delineate a physiological range outside of which an ectotherm is constrained against displaying size plasticity in response to temperature. This thermal range concept has important implications for general size-temperature studies. Furthermore, the concept of 'operating thermal conditions' may provide a new approach to (i) defining criteria required for investigating and interpreting temperature effects, and (ii) providing a novel interpretation for many cases in which species do not conform to the TSR.

Reference materials play an important part in the quality control of measurements. Rapid development of such new scientific disciplines as proteomics, metabolomics, and genomics also necessitates development of new reference materials. This is a great challenge due to the complexity of the production of new reference materials and difficulties associated with achieving their homogeneity and stability. CRMs of tissue are of particular importance. They can be counted among the matrices that are most complex and time consuming in preparation. Tissue is the place of transformation and accumulation of many substances (e.g., metabolites, which are intermediate or end products resulting from metabolic processes). Trace amounts of many substances in tissues must be determined with adequate precision and accuracy. To meet the needs stemming from research and from problems and challenges faced by chemists, analysts, and toxicologists, the number of certified reference materials should be continuously increased.

Temperature-Size Rule (TSR) is a phenotypic body size response of ectotherms to changing temperature. It is known from the laboratory studies, but seasonal patterns in the field were not studied so far. We examined the body size changes in time of rotifers inhabiting activated sludge. We hypothesize that temperature is the most influencing parameter in sludge environment, leading sludge rotifers to seasonally change their body size according to TSR, and that oxygen content also induces the size response. The presence of TSR in Lecane inermis rotifer was tested in a laboratory study with two temperature and two food-type treatments. The effect of interaction between temperature and food was significant; L. inermis followed TSR in one food type only. The seasonal variability in the body sizes of the rotifers L. inermis and Cephalodella gracilis was estimated by monthly sampling and analyzed by multiple regression, in relation to the sludge parameters selected as the most influential by multivariate analysis, and predicted to alter rotifer body size (temperature and oxygen). L. inermis varied significantly in size throughout the year, and this variability is explained by temperature as predicted by the TSR, but not by oxygen availability. C. gracilis also varied in size, though this variability was explained by both temperature and oxygen. We suggest that sludge age acts as a mortality factor in activated sludge. It may have a seasonal effect on the body size of L. inermis and modify a possible effect of oxygen. Activated sludge habitat is driven by both biological processes and human regulation, yet its resident organisms follow general evolutionary rule as they do in other biological systems. The interspecific response patterns differ, revealing the importance of taking species-specific properties into account. Our findings are applicable to sludge properties enhancement through optimizing the conditions for its biological component.
